ABSTRACT Investigation of the Therapeutic Effects of Plantago lancelota Extract and Luteolin-7-B-D-Glycoside Against DNA Damage on Human Lymphocyte and Hep-G2 Cells Aim: Plantago lanceolata (P. lanceolata) is used a phytotherapeutic plant consumed to treat or avoid of symptoms of various diseases. Luteolin-7-B-D-glycoside (LDG) is a flavoanoid which is known for its antioxidant, antigenotoxic and anticarcinogenic properties and is one of the most important components of P. lanceolata. This study aimed to investigate the possible antigenotoxic effects of the methanol extract of P. lanceolata and LDG on human lymphocyte cells and Hep-G2 cells. Materials and Methods: This study were used human lymphocyte cells and Hep-G2 cells. The DNA damaged and undamaged cells were treated with 200 and 500 μg/ml of P. lanceolata methanol extract and 10, 20 and 30 μM doses of LDG and examined sister chromatid exchange test (SCE) and antigenotoxic effect. Results: Among the study groups, only high dose of plant extract increased the frequency of SCE. In lymphocyte cells only the highest rates of P. lanceolata extract and LDG have a effect by increasing the parameters of the comet, whereas in DNA damaged cells, all test applications application doses have a reducing effect on genotoxicity. In HepG2 cells, all concentrations of both plant extract and LDG showed a genotoxic effect by increasing the formation of comet, whereas in DNA damaged cells, the minimum concentration of plant extract groups and LDG were found to reduce genetic damage. Conclusion: P. lanceolata and LDG have anti-genotoxic effect at low concentrations and genotoxic effect at high concentrations in human lymphocyte cells, while genotoxic effect at all concentrations in HepG2 cells. We can say P. langeolata plant and LDG that it is antigenotoxic effect showing in non-damage cells dose dependent, while anticarcinogenic effect showing in non-damage cancer cells.
