Determination of allele frequency of 15 STR loci in Elaziğ districts of the Eastern Anatolia region

Abstract (EN)

STR loci consist of consecutive repeated units that are length of 2-7 base pair and have particular base sequence. Variations in the number of repeated units of STR are used as marker at many genetic studies.STR loci such as D8S1179, D21S11, D7S820, CSF1PO, D3S1358, THO1, D13S317, D16S539, D2S1338, D19S433, VWA, TPOX, D18S51, D5S818 and FGA show a high degree of polymorphism in many aspect such as forensic science, paternity testing, genom mapping and population studies. 200 individuals were based in our study and 200 individuals (from indigeneous people population in Elazığ), who are healty and unrelated. They were informed and with their conset, we got their swap and blood samples for DNA products have been replicated. Using AmpFISTR Identifiller PCR Amplification Kits and AmpFISTR SE filler PCR Amplification Kits. Calculating Arlequin V.3.11 and PowerStats V.1.2 too the genetypes of 15 STR loci in Elazığ province population were determined.Each loci was reviewed one by one and allele frequence of all genotype data, heterozygosity ve homozygosity rations, power of exclusion (PE), power of discrimination (PD), probability of matching (PM), polymorphic information content (PIC), typical paternity index (TPI) values were compared with studies that perform in Turkey and significantly diffirences weren?t observed. Exclusion power was determined between 0,362-0,785, combined exclusion power was estimated as the 0,9999995. Discrimination power was observed between 0,835-0,967 and combined discrimination power was estimated as the 0,9999999999999998. Matching probability was calculated as one in 1.364x10?17 for 15 STR loci. Discrimination power is high can be said in terms of all loci. Obsorved heterozygosity and expected heterozygosity, P values were performed. Compatible or not to Hardy-Weinberg Equlibrium (HWE). Stability and compared with datas that were studied earlier in population in Turkey.Results were assessed, similarities and differences were given as statistical tables. According to HWE of compability of allel frequencies were checked. Taking into account the Fisher?s exact test P-value (P<0,05) D2S1338 (0.03226) loci P<0.05 was smaller than. However, Bonferroni?s correction (0,05/15=0,00333) was made for this locus. In addition, HWE was achieved in all other loci.Keywords: STR loci, Forensic DNA, Human Identification, DNA typing
