Research of contracting and relaxing responces of doxazosin and vardenafil on ureter smooth in model of tissue bath

Symptomatic ureterolithiasis occupy an important place in daily emergency urological practice. Ureteral colic pain is the most common symptom of the patients who have ureterolithiasis. The standart therapy includes oral hydration and nonsteroidal anti-inflammatory drugs. Ureteral relaxation and the high hydrostatic pressure in ureteral lumen facilitate dropping of stone. Some drugs relax the ureteral smooth and they are used as experimental in medical expulsive therapy. In this study that was investigated the relaxing effect of doxazosin and vardenafil on ureteral smooth in the model of tissue bath.In the study 15 New Zelland type adult male rabbits were used 8-10 mm tissue from distal and middle of ureter was hung as vertical in tissue bath. Ureters were contracted with KCL. Contracted ureters were applied 10 mM doxazosin and 10 mM vardenafil individual and together and relaxing responses were recorded.In middle and distal ureter segment after doxazosin applied was observed significant relaxation. The results are meanigful in statistically (respectively p<0,0001, p<0,0001 ).In middle and distal ureter segment after vardenafil applied was observed significant relaxation. The results are meanigful in statistically (respectively p<0,0001, p<0,0001 ).In middle and distal ureter segment after doxazosin and vardenafil together applied was observed significant relaxation. The results are meanigful in statistically (respectively p<0,0001, p<0,0001).Finally, we observed that in tissue bath model, doxazosin and vardenafil produce a relaxation effect in middle and distal ureteral segment. These data shows that doxazosin and vardenafil can be used for the medical expulsive therapy of ureterolithiasis.Keywords: Ureter smooth, doxazosin, vardenafil, relaxing
