Abstract (EN)
Tomato is the most common vegetable species of the Solanaceae family both the world and in Turkey. There are many diseases and pests adversely affecting tomato cultivation. The study carried out at the Aegean Agricultural Research Institute between 2012 and 2015 in order to develop tomato lines resistant to Tomato spotted wilt virüs, (TSWV) and knot-root nematode, a soilborne pest. For this purpose, TSWV resistant LA3667 and nematode resistant LA2941 tomato lines obtained from Tomato Genetic Resource Center (TGRC) were crossed with the fresh-market tomato line M68 in the tomato gene pool of the Aegean Agricultural Research Institute. Classical PCR, real-time PCR and biological testing methods were used for determination of durability of the resistance. Two populations have been established to develop lines resistant to TSWV and nematode. The first population was established for resistance to TSWV, and in the F2 population, 35 homozygous resistant, 74 heterozygous resistant and 41 susceptible individuals were obtained in 150 F2 plants according to the monogenic (1: 2: 1) expression. Population 2 established, in order to obtain pyrimading to two (Sw-5 and Mi genes) homozygous genes in same individual. Population 2 carried out an expansion suitable for the 9: 3: 3: 1 segregation of 92 individuals was determined in the molecular test performed at the F2 population. Five genotypes with homozygous resistance were identified in the nematode resistance gene Mi and TSWV resistance gene Sw-5. In population 1, at the F4 stage, three homozygous lines were taken for biological testing to TSWV. Results of biological test and molecular test were concordantly.
