Abstract (EN)

Alzheimer's disease; It is a medical condition that develops with memory loss, dementia (dementia) and decreased cognitive functions due to the death of brain cells over time. The blood-brain barrier is one of the most important obstacles in Alzheimer's treatment. Donepezil is among the four cholinesterase inhibitors approved by the FDA (Food and Drug Administration) in the treatment of Alzheimer's disease so far. In this thesis study, donepezil HCl was chosen as a suitable agent for our nanoparticle approach in Alzheimer's treatment due to its high selectivity to central nervous system acetylcholinesterases and low side effects. Donepezil HCl loaded PLGA nanoparticles were prepared by solvent evaporation technique. More than one method has been tried for nanoparticle preparation and the effectiveness of these methods in preparing nanoparticles has been observed. Appropriately, the I-DNP formulation was selected and the parameters affecting the particle size were examined. Encapsulation has been proven by DSC, FT-IR and 1H-NMR analyses. As a result, it was determined by in vivo CAM experiments that the I-DNP coded formulation weakens (reduces/inhibits) angiogenesis, and it was decided that the prepared nanoparticle system could be a new anti-angiogenic treatment by stimulating the cholinergic system.
