Effect of growing conditions of donor plants and different incubation temperatures on haploid plant regeneration from anthers of bread wheat

Abstract (EN)

This study was conducted to determine the effects of genotype, growing conditions of donorplants and different incubation temperatures on haploid plant regeneration from anthers of breadwheat ( Triticum aestivum L).In the study, anthers of three different cultivars (Seri 82, Genç 99, Balatilla) of bread wheatgrown under field or greenhouse conditions were cultured on P2 medium at different culturetemperatures (25 ºC, 27 ºC, 30 ºC, 32 ºC). In the experiment, percentage of anthers produced callusor embryoid, numbers of regenerated plants, green plants as well as albino plants per 100 antherscultured were determined.Result of the study showed that growing conditions of donor plants did not affect statisticallythe percentage of anthers produced calli or embryoids. The percentage of responding anthers variedbetween 0.4 % and 2.4 % depending on the genotypes and cultivar Seri 82 showed higher percentageof responding anthers than those of other two cultivars studied. Increasing culture temperature from 25ºC to 30 ºC increased the percentage of responding anthers, and the anthers cultured at 30 ºC showedstatistically significant higher percentage of response than those cultured at 25 ºC. The antherscultured at 32 ºC showed statistically significant lower percentage of response than those cultured at27 ºC and 30 ºC.Numbers of regenerated plants, regenerated green plants and regenerated albino plants per 100anthers cultured were not statistically significant influenced by the growing conditions of donor plantsand the culture temperature of anthers. The above mentioned characters of wheat anther cultureshowed statistically significant differences depending on the genotypes and cultivar Seri 82 showedhigher performance than the other two cultivars studied in those characters.From the result of the study, it was concluded that the genotype was most important factoraffecting haploid plant regeneration from anthers of bread wheat.KeyWords: wheat, genotype, donor , anther, temperature
