Abstract (EN)

ABSTRACT The Investigation of the Effects of DPPL2 Protein on Apoptosis in Breast Cancer Cells Aim: It is not known how DPPL2 protein, which is reported that its expression is increased only in breast cancer tissues in literature, performs its functions. The aim of the study is to investigate the effect of DPPL2 protein on cell death, cell cycle, and cell proliferation in breast cancer cells and to determine the role of DPPL2 protein in the cancer formation process. Material and Method: In the study, MCF10A normal mammary epithelial cells, MDA-MB 231 and MCF-7 cell lines were used. The Western Blot technique was used to analyze the presence of DPPL2 protein in cells. Suppression of DPPL2 protein was achieved by generated siRNA systems. Cell proliferation was determined by the MTS method. Cell cycle and cell apoptosis were performed by flow cytometry method. Findings: In consequence of Western Blot analysis, it was seen that DPLL2 protein was expressed in high levels in MCF10A and MCF-7, in low level in MDA-MB-231. It was seen that DPLL2 proliferation was decreased in all cell lines after the treatment of siRNA. DPPL2 cell proliferation was reduced by 20% in MDA-MB-231 and MCF-7 cells and by 25% in MCF10A cells. The amounts of DPPL2 siRNA apoptosis were determined to be 1.6% in MDA-MB-231 cells, 0.06% in MCF-7 cells and 1.98% in MCF10A cells. Conclusion: In the study, DPPL2 protein analysis was performed in breast cancer cells and mammary epithelial cells in vitro. It was seen that DPPL2 protein suppression effected in a decreased manner on cell proliferation, and in an enhanced manner on cell apoptosis. Key Words: DPPL2, Breast Cancer, epithelial cell
