Determination of myonectin expression in liver, kidney and skeletal muscle using immunohistochemical and ELISA method in trained with energy drink supplementation rats

Abstract (EN)
Energy drinks are consumed in an unbelievable way all over the world. However, the effects of energy drinks on the biological system have not been fully revealed yet. Therefore, in this study, in addition to revealing the effect of energy drink (Red Bull) on myonectin in liver, kidney and skeletal muscle tissues, it is aimed to reveal whether liver and kidney tissues synthesize myonectin or not. Twenty-eight, four-week-old male Wistar-Albino rats were included in the study. The control group (n=7), in which no application was made before the study; the exercise group (n=7) in which exercise was performed on the treadmill for four weeks (5 sessions per week/30 min at 45 cm/sec); the energy drink group (n=7) given a single daily dose of energy drink (3.57ml/kg/day) by gavage for four weeks; Four groups were randomly divided into four groups as exercise+energy drink group (n=7), in which a single dose of energy drink (3.57 ml/kg/day) was exercised by gavage for four weeks (5 sessions per week/30 minutes with 45 cm/sec). Left. The experiment lasted for four weeks and the rats were fed standard chow. At the end of the experiment, the animals were sacrificed. Myonectin amounts in tissue homogenates were analyzed by ELISA method and immunohistochemical method. SPSS 22 package program was used for data analysis. The One Way Anova test was used considering the normality criterion, and the LDS and Scheffe tests, which are Post-Hoc tests, were used to determine between which groups the differences occurred between the groups. Significance level was accepted as p<0.05. When the control groups of liver, kidney and skeletal muscle tissues were compared, the highest myonectin level was observed in the form of kidney, skeletal muscle and liver. Energy drink supplementation caused a decrease in myonectin in liver tissue and an increase in kidney and skeletal muscle tissues. Exercise caused a decrease in myonectin in liver tissue and an increase in kidney and skeletal muscle tissues. Exercise + energy drink application caused a decrease in myonectin in liver tissue and an increase in kidney and skeletal muscle tissues. Since myonectin is secreted from muscles, it is predicted that energy drink has a role in compensating for metabolic disorders that may occur during exercise. It will be useful to confirm the results with future experiments.
