Effect of melatonin and niacin on antioxidant system and lipit peroxidation at exercised rats

Abstract (EN)

In this study, it was aimed to investigate the effect of melatonin and niacin supplementation on lipit peroxidation and antioxidant system in rats, where oxidative stress was induced by exercise. Niacin and melatonin were given to forty rats except the control group, which were randomly divided into 4 groups, followed by under went treadmill exercise for 10 days. Intracardiac blood was taken after anesthesia with ketamine and Xylazine. Spectrophotometric determination of Super Oxide Dismutase(SOD), Malondialdehyde(MDA), Total Antioxidant Level(TAS), Total Oxidant Level(TOS), Catalase, Melatonin, Glutathione Peroxidase(GPX), Glutathione S-transferase(GST) levels were performed. The group given melatonin gave lower MDA concentrations in comparison to other group; however, group given (niacin + melatonin) gave higher MDA levels in comparison to control group. SOD levels were higher for the group given melatonin in comparison to other group while the group given (niacin + melatonin) gave low SOD levels in comparison to another group. Catalase levels were higher for the group given melatonin in comparison to group given niacin. On the other hand, in the control group, there was no significant change in comparison to the other groups. GPX levels: The control group was lower in comparison to the other groups. GST levels: The control group was higher in comparison to the other groups, and the group given (niacin + melatonin) gave lower levels in comparison to group given niacin. xv TOS levels: The control group was found to be lower than the group given niacin. TAS levels; The control group gave lower levels in comparison to other group, and the group given (niacin + melatonin) gave lower levels in comparison to group given niacin and melatonin. The results of this study showed that administration of melatonin to exercising rats resulted in increases of the antioxidant activity (SOD, GPX, TAS, melatonin) in comparison to control group, and it triggered more antioxidant effect in comparison to administration of niacin. Increased MDA levels that was increased with exercise of the rats while it decreased thanks to the effect of melatonin. Co-administration of melatonin and niacin did not have any synergistic effect on all the antioxidant parameters. It was determined that parameters showing that oxidation (MDA, TOS) was increased while GST, Catalase and SOD activities decreased. This shows that these enzymes are used to prevent oxidation, in accordance with previous studies. Further studies are needed by using different antioxidant molecules and trying different types of exercising models. Keywords: Antioxidant, Exercise, Lipit Peroxidation, Melatonin, Niacin.
