Abstract (EN)
Cancer-related deaths, one of the most important health problems of our time, are increasing day by day. It is known chemotherapy drugs cause side effects while destroying cancer cells. Today, to increase the effectiveness of chemotherapy drugs and reduce their side effects, there has been an increased interest in studies on the combination of these drugs with natural substances. In our study, in vitro anticancer activities on MCF-7 breast cancer and A549 lung cancer were investigated by preparing combinations of Elagic acid, a phenolic acid, with Docetaxel and Mitoxantrone chemotherapy drugs. The compusyn program was used to determine concentrations. Cell death rate was determined by apoptosis test. Antioxidant enzyme activities (CAT, SOD, GR and GST) and carbonic anhydrase enzyme activities were determined in the cell lines where the most effective doses were applied. Additionally, antioxidant and antiradical properties of substances and combinations were investigated and their effects on CAI, II and IX enzymes were investigated. Lastly, It was seen that the combination concentrations of Elagic acid + Mitoxantrone (25:1µM) applied to the A549 cancer cell line and Elagic acid + Docetaxel (50:200µM) applied to the MCF7 cancer cell line were the most effective doses, they induced apoptosis in anticancer studies, had synergistic effects and antioxidant enzyme activity results supported them. In studies outside the cell line, the combinations have antioxidant activities and inhibition effects on CA I, II and IX enzymes. Consequently, the results obtained support the suitability of the use of natural substances with chemotherapy drugs in many ways.
