2. ABSTRACT Atomic Force Microscopy Investigation of Surface Roughness Caused by Different Irrigating Agents Used in Endodontics on Biodentine and NeoPUTTY MTA Used as Perforation Repair Materials The success of root canal treatment depends on the preparation of the canals, adequate irrigation and three-dimensional filling of the root canal. The aim of this study was to investigate the effect of different chelation solutions on the surface roughness of NeoPUTTY MTA and Biodentine materials. In this study, 32 teflon blocks were used and standard grooves with a depth of 2 mm and a diameter of 2 mm were formed in the centre of the teflon blocks. The prepared blocks were divided into M (NeoPutty MTA) and B (Biodentine) groups, and these two groups were further divided into maleic acid and EDTA groups. NeoPutty (Nusmile Inc., Houston, TX; USA) and Biodentine (Septodent, Niederkasssel, Germany) were placed on Teflon blocks and condensed. The surface roughness of all samples was determined by AFM (XE-100, Park System) before soaking in 7% maleic acid (Merck Millipore, GERMANY) and 17% EDTA (Imicryl, Konya, Turkey) solutions. Afterwards, MTA and Biodentine samples were divided into two groups and the final surface roughness was determined by scanning with AFM device after being kept in maleic acid and EDTA solutions for one minute. In order to test whether there was a statistically significant difference between the surface roughness of the samples exposed to chelation solutions before and after irrigation, Mann-Withney U test was used for the comparison of two independent groups and Wilcoxon signed-rank test for dependent groups. The significance level was accepted as 0.05. According to the findings of the study, there was no significant difference between the initial roughness of NeoPUTTY MTA and Biodentine materials. However, EDTA solution produced statistically significant surface roughness in Biodentine material, while maleic acid solution produced statistically significant surface roughness in NeoPUTTY MTA material.
