In this study, antibacterial activity of different irridation times with a LED device during Antimicrobial Photodynamic Therapy was compared with each other and with sodium hypochlorite application in the teeth that were infected with Enterococcus faecalis under in vitro conditions. 106 single-rooted, extracted human teeth were used in this study. All the teeth were enlarged using chemo-mechanical shaping method up to apical size ISO 40 and sterilized. All the teeth were randomly divided into 9 experimental groups, each having 10 teeth, and 2 control groups, each having 8 teeth. Teeth in experimental groups and positive control group were infected with Enterococcus faealis ATCC 29212. Experimental groups were formed as follows: only TBO application, sterile salin+light application, PDT application for 30 seconds, PDT application for 60 seconds, PDT application for 120 seconds, NaOCl+PDT application for 30 seconds, NaOCl+PDT application for 60 seconds, NaOCl+PDT application for 120 seconds, only NaOCl application. Microbiological samples that were collected from the root canals with sterile paper points inoculated in MSB Agar broth and bacterial colonies were calculated in CFU. Results were evaluated statistically. The number of E. faecalis decreased in different ratios in all experimental groups and it was found to be significantly different from the positive control group (p<0.05). PDT application was found to be inadequate in terms of antibacterial efficacy when compared to NaOCl, and the increase in time did not appear to add any additional benefit to the antimicrobial efficacy of PDT.
