Evaluation of MMP-2, Survivin and PTEN expressions in endometrial carcinomas

Aim and hypothesis: In this study, both in endometrioid and non-endometriod adenocarcinomas, we aimed to search for the relation of expression of MMP-2, survivin, PTEN with the prognostic factors recurrence of vagen cuff disease free 5-year survival.Method: In 9 Eylul University Hospital, between years of 1984 - 2006, biopsies of 95 cases who had underwent either a radical operation for endometrial carcinoma or a curettage for diagnostic purpose were investigated for the expression of MMP-2, survivin, PTEN. The expression results of these antibodies were evaluated for their relation with the stage, histological/nuclear grade and disease free 5-year survival.Results: 21 (22.1%) of the cases with endometrial carcinoma had type 2 endometrial carcinoma, 74 (77.9 %) of them had type 1 endometrial carcinoma. In the present study, the mean age of the cases is 61 (median age is 70.0 -+10.3). The 76.8% of the cases are stage 1, the rest of them are stage 2 and 3. We couldn?t find a significant relationship between the cytoplasmic immunoreactivity of the MMP-2 and the stage, histological, nuclear grade of the tumor. In our study a significant relation is found between the nuclear expression of survivin and the nuclear, histological grade. Survivin is determined to be expressed in a higher ratio in endometriod adenocarcinomas, than the nonendometrioid adenocarcinomas. It is found that PTEN expression in advanced stage tumors is low. MMP-2, survivin and PTEN determined to have no relationship with 5-year disease free survival, overall survival and with recurrence of vagen cuff.Conclusion: In this study use of survivin, MMP-2 and PTEN in endometrium carcinomas determined to have no relationship with 5-year disease free survival, overall survival and with recurrence of vagen cuff, so their usage in routine is controversial. Therefore studies should be done evaluating different prognostic factors in large series with long term follow-up.Key words: Endometrial carcinoma, prognosis, survival rate, PTEN, survivin, MMP-2.
