Evaluation of Trp64Arg gene levels and tumour necrosis factor-related proteins in preterm birth pregnant

Abstract (EN)

Aim: This study aimed to compare β3-AR gene expression and Trp64Arg frequency in PTB with healthy pregnant and to determine the relationship of gene levels with CTRPs and plasma lipids, and to investigate pathological changes in placental tissues. Material and method: CTRP levels and plasma lipid levels were determined by the ELISA method in blood samples taken from 40 pregnant diagnosed with PTB and 40 control who completed the normal pregnancy period. Gene expression was evaluated by RT-PCR method. Pathological changes were examined using hematoxylin-eosin staining. Results: In the study group, β3-AR gene levels significantly decreased and Trp64Arg gene frequency was significantly increased compared with the control. While CTRP3 and TC levels decreased in the study group, CTRP4 and LDL levels increased significantly. ROC analysis revealed that LDL, HDL, CTRP3, and CTRP4 levels could be predictive biomarkers for PTB. On pathological examination of placenta, hydropic degeneration, perivillous and intervillous fibrin deposition were significantly higher in the study group than in the control group. Fibrin levels increased in the study group and were found to be associated with birth week and CTRP4. Conclusion: The relationship between CTRP levels and PTB was investigated for the first time. CTRPs were associated with blood lipid levels, especially HDL and LDL levels. While CTRP3 decreases as a result of PTB complications, CTRP4 increases as a defense mechanism. Decreased β3-AR levels and increased Trp64Arg frequency in the placenta may have caused fibrin accumulation by reducing placental perfusion, which may have contributed to the pathophysiology of PTB.
