Abstract (EN)

Rhabdomyolysis due to ischemia-reperfusion(I-R) in the lower extremity is a pathological condition that can develop in some surgical cases. Ethyl pyruvate(EP) has been shown to have a protective and therapeutic effect against I-R injury by studies. The objective of our study was to evaluate histologically the effect of EP on local and remote organ injury resulting from the I-R in the lower extremity. In this study, 28(4 group; n=7) C57BL/6 male mice were used. The I-R model [ischemia(3h)-reperfusion(3h)] was performed by placing the bilateral tourniquet on the hip-joint of the mice. One of the experimental groups was administered 150 mg/kg EP intraperitoneally 15 minutes before ischemia(Grup-3) and the other at the onset of reperfusion(Grup-4). Lung, kidney and gastrocnemius muscle tissues were examined histopathologically. Muscle: H-E;when Group-3 tissues were examined, a small amount of myofibrillary disorganization and degeneration was observed compared to Group-2, while Group-4 tissues showed less myofibrillary disorganization and degeneration than Group-3. TUNNEL;less TUNEL-positive cells were counted in Group-4 tissues compared to Group-3 tissues. Lung: H-E;mild mononuclear cell infiltration was seen in Group-3 tissues. Mononuclear cells in the bronchiolar submucosa were less seen in the lung tissues of Group-4, and it was found to be similar to Group-1. TUNEL;less TUNEL-positive cells were counted in Group-4 tissues compared to Group-3 tissues. Kidney: H-E;Erythrocyte extravasation and tubular dilatation in the tissues of the Grup-3, tubular dilatation and erythrocyte extravasation were observed in some places in the tissues of the Grup-4. But when compared, it was found that the protective effect of EP was more effective at the beginning of reperfusion. TUNEL;less TUNEL-positive cells were counted in Group-4 tissues compared to Group-3 tissues. As a result of this study;EP has a protective effect on I-R injury that varies depending on the time of administration.
