Keratinocytes differentiated from mouse embryonic stem cells effect of wound healing in mouse that single dose of steroid injected

In today's world, chronic systemic and local factors depending on increase of chronic drug use, cause impaired wound healing. Also frequency of chronic non-healing wounds is getting higher. The aim of this thesis is to analyze the effect of embryonic stem cellsdifferentiated keratinocytesto wound healing in the use of steroid use which impairs wound healing. In this study, keratinocytes was obtained from mouse embryonic stem cells by immunohistochemistry study. In total, 64 mice were divided into 4 equal groups. These groups were; only surgical wounds group; steroid given wound before the creation of the surgical wound + steroid group;embryonic stem cellsdifferentiatedkeratinocytes implemented wound after the surgical wound created + cell group and the last one is surgical wounds that are steroid administered previously, then after surgical wound stem cellsdifferentiated keratinocytes transferred to this wound + steroids + cell group. After creation of wounds, wound healing processes followed by taking a biopsy from subjects in 1, 5, 10, and 21 days and these groups were compared with each other. The biopsy samples was evaluated in terms of H & E stain and distributions of MMP2 , MMP9 , IL1 β , TNFa , MCP1 , HIF1α , FGF2 , EGF, COL1, IL10. In the result of our study we conducted that: application of steroids adversely affect wound healing, wound healing affected positively in the groups which we applied embryonic stem cells differentiated keratinocytes compared to the group without keratinocyte application.
