Investigetion of the effects of experimental amyloidosis in mice on the immun system

Abstract (EN)
In this study, investigation of the effects of experimental amyloidosis in mice on the immune system was aimed. 120 female mice 7 weeks old from "Swiss albino" race used in the study were divided into six equal and 20 animals in each groups. To induce amyloidosis Complete Freund's Adjuvant - Casein emulsion was injected to experimental group and treatment group as intraperitoneally and subcutaneously during the 8th and 10th weeks of the study. Additionally two doses of methylprednisolone was injected into the left hind leg of the mice in the treatment group intramuscularly. Following the injection at the end of the 8th week blood samples from mice were taken and their necropsy was examined. At necropsy examination, the liver, spleen and kidney tissues were taken and the presence of amyloid was defined by congo red while the presence of Interleukin (IL)-1β, IL-6, TNF-α and Amyloid A were defined by immunohistochemistry technique. SAA, IL-1β, IL- 6 and TNF-α were determined by ELISA technique. In sera the highest rate of serum amyloid A (SAA) in the treatment group was (Group-5) (p<0,05) and in the experimental group was (Group-3), and in the tissue the most severe amyloid deposition was detected at Group-3. The highest rate of TNF-α , IL-1β, IL-6 in sera was detected at Group-3 (p<0,05). Consequently it is possible to assert that SAA , IL-1β, IL-6 and TNF-α play a major role in the formation of amyloidosis, and even it can be claimed that there are significant correlations between SAA and proinflammatory cytokines and between IL-1β and IL-6. Also it is concluded that as for suppressing amyloidosis, it can be suitable to give repetitive and different doses of methylprednisolone.
