Abstract (EN)

ACE2- protein is an 805 amino acid, type I integral membrane protein belonging to the angiotensin-converting enzyme family of dipeptidyl carboxydipeptidases and is encoded by the ace2 gene located at position Xp22.2 on the X chromosome. The protein has an extracellular domain, a single transmembrane domain and a cytoplasmic tail. ACE-2 is expressed in many tissues in the human body and is also found in mice, ferrets and golden hamsters. ACE-2 is the cell entry receptor of SARS-CoV-2. The virus initiates membrane fission by binding to ACE-2 through the receptor binding domain (RBD) of the spike protein. The Vero cell is a continuous cell line prepared from renal epithelial cells of the African green monkey and is capable of continuous proliferation. They mainly contain a deletion in their type I interferon gene, so they cannot produce interferon alpha or beta, making them more susceptible to viral infection. For this reason, they have also been used to study a number of viruses and in the preparation of human vaccines. In this thesis, the host cell (Vero) used for COVID-19 vaccine production was grown in stationary culture and microcarrier suspension culture. At the end of production, the production models of ace2 expression, which is the virus binding receptor, were compared. When the findings were examined, the presence of ACE-2 receptor in Vero cells produced in both cultures was confirmed by conventional PCR analyses. Similar results were obtained when RT- PCR results were calculated with ΔΔCT.
