Evaluation of Total Antioxidant - Oxidant Status and Gene Expression Values of Various Pulp Capping Materials on Human Dental Pulp Stem Cells Aim: The aim of this study was to evaluate the effects of various pulp capping materials (ProRoot® MTA, Biodentine™, Pulpine Mineral, Endo Repair and ZnOEu) on human dental pulp stem cells with TAS-TOS analyzes and gene expression values remarking TGF-β1, BMP-2, ALP and Bcl-2 biomarkers. Materials and Methods: In our study, American Type Culture Collection human dental pulp stem cells were used. The filter diffusion test was applied in accordance with ISO standards. At the end of the 24th and 72nd hours; after TAS and TOS biochemical analysis, gene expression levels were measured by qRT-PCR method (n=3). Kruskal Wallis Variance Analysis and Mann-Whitney U test were used for independent group comparisons between the measurements. Results: During the initial 24 hours, we observed that only the ProRoot® MTA group had a high antioxidative defensive mechanism. We determined that the antioxidative defense level had been increased in all groups with the highest value in BiodentineTM, Pulpine Mineral groups while the lowest antioxidative defense level seen in ZnOEu group in between the 24-72nd hours (p<0.05). TGF β1, BMP-2 and ALP gene expressions were found to be more frequent in Biodentine™ and Endo Repair materials in relative to the other materials. (p<0.05). It was observed that Bcl-2 gene expression levels decreased in all materials at the end of 72nd hours. Endo Repair and ProRoot® MTA groups performed overexpression in comparison to the control group after 72 hours (p<0.05). Conclusion: According to the TAS and TOS values, it was determined that all materials have an oxidative effect on pulp and act antioxidant defense mechanism. We also found that Biodentine™ and Endo Repair groups had good bioactivity as well as the biocompatibility.
