ABSTRACTMSc ThesisA STUDY ON POSSIBLE MUTAGENICITY OF DIFFERENT TYPES OF PEST C DES BYAmes/Salmonella/Microsome TEST SYSTEMAhmet BARIŞAfyon Kocatepe UniversityGraduate School of Natural and Applied SciencesDepartment of BiologySupervisor: Prof.Dr. Muhsin KONUKIn this study, the mutagenic effect of 4 different pesticides (piperophos, fenchlorphos, dioxacarb, promecarb) used in agriculture widely have been investigated by using short timebacterial mutagenicity test system namely Ames/Salmonella/Microsome test. In this study,Salmonella typhimurium TA 98 ve TA 100 strains were employed. Therefore, both test strainswere tested in the absence or presence of S9 metabolic activation. For piperophos, 10 µl/plate,1 µl/plate, 0,1 µl/plate, 0,01 µl/plate and 0,001 µl/plate concentrations and for other testcompounds 100 µg/plate, 10 µg/plate, 1 µg/plate, 0,1 µg/plate and 0,01 µg/plate concentrationswere investigated in two paralel independent experiments. At the absence and presence of S9,sodium azid was used as a positive control for TA 100. At the absence of S9, 4-nitro-o-fenilendiamine, at the presence of S9, 2-aminofluorene was used as a positive control for TA98. DMSO solution and spontaneous control groups were used as a negative control. Results ofexperiments were compared with positive and negative control groups data. Consequently;among these four pesticides, only 0,01 and 0,001 µl/plate doses of piperophos was foundmutagenic in TA 98 with S9.Key words: Ames test, mutagenicity, mutajenite, piperophos, fenchlorphos, dioxacarb, promecarbv
