Abstract (EN)

Induced pluripotent stem cells (IPSC) are pluripotent cells that obtained from adult somatic cells by genetic reprogramming in vitro via transcription factors. Due to its pluripotency, it has the ability to differentiate into almost many cell types. The use of the IPSc cell source for regenerative purpose removes ethical problems about embryonic stem cells. In this study, pluripotent stem cells were obtained from fibroblast cells via for main transcription factors. The plasmid containing green fluorescent protein (GFP) gene with Oct4, Sox2, Klf4 and v-Myc transcription factors was amplified in E. coli JM109 strain and purified. Fibroblast cells were transfected with plazmid by using a lipid-based nano vehicle. Characterization studies of octadecylamine-based cationic lipid nanoparticle (OLN) synthesized in our laboratory were performed by zeta-sizer. The cytotoxicity of the nanoparticle was determined by MTT assay (ISO 10993-5) OLN- plasmid conjugates were transferred into the L929 cell line, and the transfection process in fibroblast cells was evaluated via screening GFP expression. Expression of the Oct4, Sox2, Klf4 and v-Myc genes in the cells were determined by the RT-qPCR. Then, the obtained UPS cells were loaded to a cellulose-based hydrogel wound dressing biomaterial which was designed in our previous studies.
