Abstract (EN)
The aim of this study was to determine genotoxic effect of flurbiprofen which is used as an active ingredient in NSAID analgesics. To determine genotoxic effects of flurbiprofen, chromosome aberration test and RAPD-PCR test were performed in vivo by using rat bone marrow cells. Also, total oxidant and total antioxidant positions were determined by spectrophotometric method. In this study, albino rats were treated with 29.25, 58.50 and 117 mg/kg flurbiprofen for 12 or 24 hours. Percentage of abnormal cells and chromosome aberrations were not statistically influenced by any concentrations and time periods. However, mitotic index significantly decreased in both 12 and 24-hour treatment period. This reduction in the 12 hour treatment period was concentration-dependent manner. Also in the highest concentration of 12 hour treatment period, reduction of mitotic index was more than in positive control. RAPD-PCR procedure was performed with 7 primers by using genomic DNA obtained from bone marrow of flurbiprofen treated rats. Then the product of RAPD-PCR was analyzed and evaluated by agarose gel electrophoresis and a significant difference was not found between the variables. A long with this result, test substance did not have a significant effect on the genomic template stability. Antioxidant values decreased in 58.50 and 117 mg/kg concentrations of flurbiprofen for 12 hour period and this increased the oxidative stress. In conclusion, this study showed that flurbiprofen significantly reduced mitotic index in rat bone marrow. I consider that this cytotoxicity may be linked with oxidative stress.
