Galangin and quercetin efects on cell viability and proliferation in neuroblastoma cells

Abstract (EN)
Objective: This study aimed to investigate the effects of galangin and quercetin on cell proliferation in SHSY-5Y neuroblastoma cancer cells. Material and Methods: In this study, the SHSY-5Y neuroblastoma cell line was propagated using cell culture techniques. Appropriate doses of galangin and quercetin were determined, and a total of four groups were established. MTT assay, clonogenicity, matrix invasion, and immunofluorescence in vitro experiments were conducted. The collected data were analyzed using the SPSS program. Results: According to MTT results, appropriate doses were determined as the combined treatment group. Considering the dose applications, quercetin 40 µM and galangin 40 µM doses were accepted as the most effective doses on cell proliferation. It was observed that it had a 50% decreasing effect on cell proliferation, therefore these doses were used in the combined groups. In the clonogenicity experiment, it was observed that clone formation was quite low in the quercetin group. In the galangin group, more cells were located in the periphery. In the combined treatment group, it was observed that the cells spread to the entire surface. In the immunofluorescence experiment, it was observed that TCF8/ZEB1 protein was effective on the expression in the galangin and quercetin applied groups compared to the control group. In the combined treatment group, where the drugs were applied together, it was found that protein expression increased compared to the groups where the drugs were applied individually. In the matrigel invasion experiment, it was observed that the effect on the cells increased depending on the increase in the drug dose. It was observed that 40 µl quercetin showed the most effect, the 40 µl galangin treatment caused a significant difference compared to the control. Conclusion: Quercetin and galangin were found to reduce cell proliferation in the SH-SY5Y cell line at the determined doses. Keywords: Galangin, Quercetin, Neuroblastoma, Cell Proliferation
