Abstract (EN)
Phenolic acids have many positive effects such as antioxidant, antimutagenic and anticancer. The benefit-harm relationship of the chemotherapeutic agents used for cancer treatment is discussed, and thus the search for new active agents continues. In our study, combinations of Gallic acid, phenolic acid, with chemotherapeutic agents such as Docetaxel and Mitoxantrone were prepared and in vitro anticancer activity on lung cancer cell line (A549) was investigated. The CompuSyn program was used to determine concentrations. Antioxidant enzyme activities (CAT, SOD, GR and GST) and carbonic anhydrase enzyme activity were determined in the cell line where the most effective dose was applied. In extracellular studies, the antioxidant and antiradical activities of substances and combinations and their effects on CAI, II and IX enzymes were investigated. Consequently, a synergistic effect was observed in the use of Gallic acid and Mitoxantrone, cell apoptosis was achieved by significantly reducing viability 87.10 % at the end of 24 hours (15.85 %) and antioxidant enzyme activity results were also supportive. In extracellular studies, it has been observed that the combinations have antioxidant and antiradical activities and inhibition effects on CAI, II and IX enzymes. When the results were evaluated, it was concluded that the combination of Gallic acid and Mitoxantrone (25 µM + 0.5 µM) might be suitable for the treatment of lung cancer. Although positive results were obtained in combination studies prepared with Docetaxel, it was thought that it is not suitable for treatment because no synergistic effect was observed.
