Abstract (EN)

Introduction and Aim: In this thesis, the results of immunochromatographic and real-time PCR methods in the diagnosis of Rotavirus, Adenovirus and Norovirus infection in Sakarya were analyzed, and the results of these tests were compared. Materials and Methods: 84 samples from patients presenting to our hospital with diarrhea complaints were included in the study. Of these, 67 were positive samples with immunochromatographic test (ICT) for Rotavirus and Adenovirus. 17 samples which were negative for Rota/Adenovirus and bacteria/parasites were taken as control group. All samples were tested using multiplex-RT-PCR method. Results: Out of 84 samples 62 were Rotavirus positive (%74) and 22 were Rotavirus negative (%26), 5 were Adenovirus positive (%6) and 79 were Adenovirus negative (%94) with the ICT method. Real-time PCR analysis of all samples; Rotavirus was found to be positive in 49 (58.5%), negative in 35 (41.5%) and Adenovirus was positive in 40 (48%), and negative in 44 (52%). Real-time PCR showed 11 (13%) positive and 73 (87%) negative results for Norovirus. There were significant, medium and low level agreement for Rotavirus and Adenovirus among ICT and PCR methods (Rotavirus-ICT/Rotavirus-PCR; r: 0.540, p <0.001, Adenovirus-ICT/Adenovirus-PCR; r: 0.264, p <0.016 ). When RT-PCR is accepted as reference method; Rotavirus ICT sensitivity; 93%, specificity; 54.3%, PPD, 74%, NPD, 76.4% and Adenovirus ICT sensitivity; 12.5%, specificity; 100%, PPD, 100%, NPD, 55.7%. Conclusion: A significant but medium and low level of compliance was found between the ICT and real-time RT-PCR methods for Rotavirus and Adenovirus. ICT; It is a test suitable for small labs, suitable for screening and cheap, fast, useful, practical, no hardware. Although real-time PCR is a more sensitive and specific method, it is expensive, slower, requires hardware and is not available in every laboratory.
