Effects of acyl and desacyl form of ghrelin hormone on hepatic steatosis and biochemical findings in rats

Abstract (EN)

This study was carried out to evaluate the effect of acyl and desacyl forms of ghrelin hormone on fatty liver and biochemical findings in rats. The study was conducted with 36 male Wistar rats. Rats were divided into 6 equal groups as control, acyl ghrelin, desacyl ghrelin, acyl/desacyl 3:1, acyl/desacyl 1:1 and acyl/desacyl 1:3. The rats were administered subcutaneous placebo or 200 ng/kg hormones twice a day for 14 days. Oral Glucose Tolerance Test (OGTT) was applied on the 15th day of the study, and the Insulin Tolerance Test (ITT) was applied on the 16th day and sacrification was performed on the 17th day. Some biochemical data in the blood of rats and diacylglycerol (DAG), glycogen, protein kinase C and PPAR-γ levels in the liver were determined. In addition, histological examinations were made in the liver tissue of rats. In the results of working; The highest plasma total cholesterol and VLDL-C levels were observed in acyl/decacyl 1:3 group, and insulin and HOMA-IR levels were lower in groups given acyl and desacyl ghrelin together (p<0.05). It was found that PPAR-γ gene expression levels increased in acyl ghrelin and acyl/desacyl 1:3 groups compared to the control group (p<0.05). The expression of the protein kinase C gene was the highest in the acyl/desacyl 1: 3 group compared to the control group (p<0.05). It was observed that the most intense degenerative findings compatible with steatosis in the liver were in the acyl ghrelin group (p<0.05). n this study, it was found that acyl / desacyl 1: 3 group increased PPAR-γ gene expression, serum total cholesterol, HDL-C and VLDL-C levels in the body, similar to the acyl ghrelin group. It has also been shown that desacyl ghrelin is more effective in regulating blood glucose level.
