The Evaluation of Killer Cell Immunoglobulin- like Receptor(Kir)GenePolymorphism in Glioblastoma Multiforme

Abstract (EN)
Purpose: Our aim was to evaluate the distribution of Kir gene polymorphism in order to predict the clinical course, enlight the genetic mechanisms and be guidance for the potential therapeutic methods. Material and Methods: Thirty-one adult patients who were admitted to our Department of Neurochirurgie and diagnosed as GBM between October 2013 and January 2014, and 50 control subjects were included for the study. After obtaining informed consents, demographical, clinical information and blood samples were collected from each participant. After isolating DNA, Kir genotyping was performed with the sequence spesific oligonucleotid probes (SSOP) method. Results: Twenty male, 11 female a total of 31 GBM patients and 32 male and 18 female, a total of 50 control subjects were included for the study. The mean age (53,5 years vs. 53.8 years, respectively) and gender distribution (male/female: % 64.5/35.5 vs. % 64/36, respectively) were comparable among patients and the controls (p>0.05). Sixteen different Kir gene including inhibitor, activator and pseudogenes were investigated for each sample and the framer genes including KIR2DL4, 3DL2, 3DL3 and 3DP1 was present in all of the patient and controls. In addition, the inhibitor KIR genes, 2DL3 gene was significantly more common in patients than controls (p<0.05). Conclusion: This study showed the inhibitor Kir gene 2DL3 as a predisposition for GBM. The identification of the potential link between GBM cells and immune system genetics is critical for predicting familiar predisposition and early diagnosis. In addition, this clue may be a key factor for developing post-surgery individual immunotherapeutic therapy models in future. Keywords: Kir gene, NK cells, Glioblastome multiforme, SSOP.
