Abstract (EN)
Identification of genes related to toxic boron stress in boron hyperaccumulator species is thought to illuminate boron tolerance mechanisms in plants. With this in mind, toxic amount boron treated G. perfoliata leaf and root cDNA libraries were created. cDNA libraries were transformed into yeast cells and screened in the presence of high concentration of H3BO3. After selecting 1 transgenic yeast plasmid among 10 boron-tolerant transformants and next generation sequencing, gene properties were annotated using BLASTX. The results show that the identified GpEEFA1a gene enhances stress signal metabolism in Saccharomyces cerevisiae. Retransformation of yeast cells with the selected plasmid provided boron tolerance in the cells. Expression levels of this stress-related gene were checked by qPCR in G. perfoliata leaves. In this study, a gene providing slightly boron tolerance was identified in G. perfoliata.
