Investigation of Cucumber mosaic virus (CMV) by taqman real-time RT-PCR method in cucurbits in Hakkari province

In this thesis study; the Cucumber mosaic virus (CMV), which is one of the viral agents causing harmful and economic losses in cucurbit cultivars grown in Hakkari province, was investigated. CMV is one of the important viruses that can infect many agricultural products. Leaf samples were collected from cucurbits, showing signs of yellowing, necrosis, mosaic, and deformity on the leaves suspected in terms of virus disease, from Kırıkdağ, Otluca, and Merzan areas during survey studies which were conducted from August to September 2022.The presence of the CMV in a total of 50 symptomatic leaf samples (including 7 watermelon (Citrullus lanatus L.), 3 melon (Cucumis melo L.), 17 cucumber (Cucumis sativus L.), 8 zucchini (Cucurbita pepo), 9 pumpkin (Cucurbita moschata), 2 gourds (Lagenaria siceraria) and 4 acrid (Cucumis melo var. Flexuosus) siceraria)) was investigated with the TaqMan real-time RT-PCR method using probes and primers specific to the 105 bp region of the coat protein (CP) gene of the virus.CMV, which was detected in 12% of 50 samples, was most commonly detected in Otluca and melon with a rate of 33.3%. This thesis is the study to investigate CMV in cucurbits with TaqMan real-time RT-PCR method.
