Investigation of ectoine production capabilities of halophilic microorganism isolates

In this study, halophilic microorganisms were isolated from water samples taken in June from the Pülümür spring salts at an altitude of 1750 m in Tunceli. Characterization of the obtained isolates based on 16S rRNA gene analysis was performed, and 2 halophilic strains were selected to be studied. Selected Halomonas sp. P-6-8-3 and Chromohalobacter israelensis Ba1 strains were examined for their ectoine production abilities. Ectoines obtained by bacterial milking (by applying hyperosmotic and hypoosmotic shock) method using two different media were analyzed by HPLC (High-Performance Liquid Chromatography) method. As a result of HPLC analysis, it was determined that both strains produced ectoine, and the concentration of ectoine produced differed between the media and between hypoosmotic shocks with solutions containing different concentrations of NaCl.
