Abstract (EN)
Precision oncology aims to find effective treatments for each patient individually. Preclinical models, such as patient-derived tumor xenografts (PDTXs), present a lot of technical issues with regard to predicting therapy responses. Patient-derived organoids (PDOs) offer promise for personalized medicine. PDOs are structures that can be obtained from individual cancer patients and can recapitulate morphological and genetic features of the original tumor among the 3D culture systems. However, non-standardized and ill-defined culture procedures are a lack of in cancer organoid research. This study aims to establish the organoids of colorectal cancer (CRC) patients by using an in-house protocol and to demonstrate histological and genetic similarities to the original tissue. After obtaining patient consent, fresh tissue resection samples (tumor and non-cancerous adjacent counterparts) were acquired from colorectal cancer patients (n=17) over the age of 18. Cells were counted and embedded in matrigel after mechanical and chemical dissociation of tissues. As a result, we were able to generate organoids from tumor biopsies and healthy tissues with over %90 successful rates. According to an independent pathology consultant, organoids have characteristics of CRC such as cribriform structure as in their original tumor samples. Finally, bioinformatics analysis demonstrated that a patient and matching disease model share similar somatic mutation profiles, particularly for mutations pertaining to well-recognized mutated genes of the tumor. Therefore, our in-house optimizations have provided us with a robust and efficient protocol for PDO establishment and made it possible to use them as a tool in personalized treatment approaches in a proper way.
