Determination of some probiotic characteristics of lactobaci?llus species orginated from animals

Abstract (EN)

In this study, 18 Lactobacillus spp. strains isolated from chicken gastrointestinal system studied probiotic characteristics such as lactic acid and hydrogen peroxide production, antimicrobial activity, antibiotic sensitivity, resistance against acidity, sensitivity against bile salts and exopolysaccharides (EPS) productions were investigated. Percentage of lactic acid produced by strains was found 0.42 to 0.90% and the amount of hydrogen peroxide production was determined between 0.18?3.77 µg/mL. In addition, EPS productions of strains were determined as a range of 10.60-180.36 mg/L. Disc diffusion method was used to examine antibiotic resistance among Lactobacillus spp. strains and strains were 100% sensitive to penicillin-G (78% moderate sensitive and 22% sensitive) and ampicillin, 100% resistant to gentamicin and kanamycin. The antimicrobial activities of Lactobacillus strains against seven different bacterias such as Shigella sonnei Mu:57, Pseudomonas aeruginosa ATCC 27853, E. coli O157:H7, Escherichia coli ATCC 11229, Salmonella enteritidis ATCC 13076, Staphylococcus aureus ATCC 25923 and Listeria monocytogenes ATCC 7644 were found to be respectively, 100, 94.5, 83.3, 77.7, 72.2, 66.6 and 5.5%. Lactobacillus strains? resistance to different pH values were determined weak. Although the grow rate was low at low pH?s such as pH 1, it was observed the strains can be still alive. Acidity ratio was increasing in the environment, the density of bacteria decreased. Bile tolerance of Lactobacillus strains changes depending on the concentration. The development of all strains was decreased by the increase of bile concentration from 0,06 to 0,30%. Autoaggregation of 5 Lactobacillus strains that have high EPS production and coaggregation abilities of E. coli ATCC 11229 and Salmonella enteritidis ATCC 13076 with that 5 strains were determined. The highest autoaggregation percentage value was 33,3% with L. acidophilus BAZ36. Considering the coaggregation percentage of E. coli ATCC 11229, L. acidophilus BAZ36 has the highest coaggregation percentage (28,3%), when the coaggregation percentage of Salmonella enteritidis ATCC 13076 considered, the highest coaggregation percentage was 25,0% with L. delbrueckii ssp. delbrueckii BAZ32.
