SUMMARYInvestigation of the EF-24?s effect on Taxol 's apoptotic and anti-proliferative response in HeLa cell lineThe molecules used as stabilizers of microtubules inhibit metaphase and anaphase transition and direct the cells to apoptosis due to the change in microtubule dynamics. Taxol is one of these molecules which is used as chemotherapeutic agent in various cancer types. Another chemotherapeutic agent used in a variety of cancer studies is EF-24, one of the curcumin?s synthetic analogues. EF-24, under in vitro conditions, is known to stimulate apoptosis of various cancer cells by inhibiting cell proliferation.In this study, we aimed to research the apoptotic or anti-proliferative effect of Taxol and EF-24 on HeLa cells , Cyclin D1 and PI3K genes which play a crucial role in cell cycle progression and the effects of caspase-3 and caspase-9 genes in charge in apoptotic pathway on the mRNA expression levels.In our study, after Taxol and EF-24 were implemented on Hela cell line both individually and combined, their cell cultures were ended in 24 and 48 hours? time. The cell viability rates were determined by MTT method. In addition, quantitative real-time PCR method was used to determine the levels of mRNA expressions genes related to cell proliferation and apoptosis of Taxol?s and EF-24's .In our study, we observed that when EF-24 and Taxol were applied separately, the viability of cells decreased in increasing concentrations. However, when these drugs were used together, it has been observed that the viability of cells decreased less-especially compared with the application of EF-24 alone- and that when they were used together they were creating an antagonistic effect. At the end of the study, it has been shown that especially EF-24 used with a high dose of Taxol decreased the effect of Taxol, EF-24 used with a low dose of Taxol increased the effect of Taxol. Similarly, according to the data obtained from mRNA analysis, as a result of the use of those drugs together, as we look to the levels of caspase-3, caspase-9, cycline D1 and PI3K mRNA, supporting symptoms have been obtained.Key Words: Gene expression, Apoptosis, Cervix Cancer, Taxol, EF-24
