Effectiveness of applying prophlactic glycyrrhiza glabra in acute respiratory disstress syndrome induced by hydrochloric acid and lipopolysacharide

Abstract (EN)

Acute Respiratory Distress Syndrome (ARDS); It is a condition characterized by bilateral diffuse alveolar damage, variable degrees of ventilation perfusion impairment, hypoxemia, decreased lung compliance, and bilateral diffuse infiltration on chest X-ray. Patients often need a mechanical ventilator. The patients have high morbidity and mortality. Early recognition of ARDS is important in terms of providing appropriate treatment and reducing morbidity and mortality. Our lungs are an organ that is exposed to high levels of oxygen radicals. The defense mechanisms of the human lung against oxidants include metal-binding proteins, mucus components and small molecular weight antioxidants, vitamins, and various enzymatic antioxidants. Antioxidant compounds are being tested in the treatment of ARDS. In our study, we aimed to evaluate the effectiveness of Glycyrrhiza Glabra pre-application. In our study, 24 wistar female rats were used. Rats were randomly divided into 4 groups. Group 1; The control group is the group in which a 1 cm skin incision was made on their neck and closed. Group 2; ARDS Group, the group in which Acute Respiratory Distress Syndrome was created by applying intratracheal Hydrochloric Acid+Lipopolysaccharide. Group 3; The group in which 100 mg/kg Glycyrrhiza Glabra extract was administered by gavage for 2 weeks and Acute Respiratory Distress Syndrome was formed. Group 4; The group in which 200 mg/kg Glycyrrhiza Glabra extract was administered by gavage for 2 weeks and Acute Respiratory Distress Syndrome was formed. The rats were anesthetized with intraperitoneal ketamine. Intracardiac blood was taken from all rats 24 hours after intratracheal Hydrochloric Acid+Lipopolysaccharide was administered to rats planned to develop Acute Respiratory Distress Syndrome and all of them were sacrificed. After the rats were sacrificed, lung resection was performed. Lung preparations were scored for total lung injury score. Serum Superoxide Dismutase, Glutathione Peroxidase, Myeloperoxidase activities, Malondialdehyde, Total Antioxidant and Total Oxidant levels from blood were studied and statistically evaluated. According to the results we obtained in our study, lung injury scores in the ARDS group were statistically significantly higher than in the control group. There was no significant difference in lung injury score in the groups treated with Glycyrrhiza Glabra extract compared to the ARDS group. Serum SOD levels were significantly higher in Group 3 compared to the control group. Serum GPX levels were significantly higher in the groups treated with Glycyrrhiza Glabra extract compared to the control group. Serum SOD and GPX levels of Glycyrrhiza Glabra extract groups did not differ significantly compared to ARDS group. MPO, MDA, TAS and TOS levels did not differ statistically between the groups. In our study, we could not show any effect of Glycyrrhiza Glabra extract application on oxidant damage in rats with ARDS. We think that the studies to be conducted with the use of separated and bioavailability proven formulas of Glycyrrhiza Glabra extract and the creation of a control group that has not been exposed to any stress factor will yield more valuable results.
