Aim: To evaluate the effectiveness of the HA-PRF membrane, a novel biomaterial, on gingiva-derived mesenchymal stem cells by comparing it with L-PRF and T-PRF membranes. Materials and Methods: Gingiva and blood samples were obtained from a systemically healthy male volunteer. Blood samples were centrifuged in glass tubes and nano-sized hydroxyapatite-coated tubes at 2700 rpm for 12 minutes, and in titanium tubes at 2700 rpm for 20 minutes. The obtained PRF clots were made into membranes in a metal box using a compressor and a metal lid. Viability, apoptosis, ELISA, and qRT-PCR experiments were performed on gingiva-derived mesenchymal stem cells, which were previously isolated and characterized, with PRF membranes on days 1, 3, and 7. The effects of the secretions of PRF membranes on days 1 and 3 on wound healing were investigated in vitro. The morphology of PRF membranes on days 1, 3, and 7 was evaluated by SEM. The data obtained as a result of the experiments were statistically analyzed. Results: In the MTT analysis, the highest average viability values were observed in the L-TZF and HA-TZF membrane-containing group on day 7 in a statistically significant manner. In the Annexin-V/PI analysis during flow cytometry, no statistically significant difference was found between the percentages of apoptosis and necrosis. In the wound healing experiment, a statistically significant smaller wound surface area was observed at 12 and 24 hours in the positive control and HA-TZF membrane-containing group. Conclusion: The HA-TZF membrane did not exhibit cytotoxic effects on gingiva-derived mesenchymal stem cells. TZF membranes demonstrated an anti-apoptotic effect against stem cells. HA-TZF membrane is more effective in wound healing compared to L-TZF and T-TZF membranes.
