Investigation of the effect of hyperthermia on the male germ cells by the structural analysis

Abstract (EN)
Hyperthermia can be caused a heat shock when the body temperature rise up 41 °C. Testicular heat lower than body heat in mammalian. Scrotal temperature increase can cause lower quality sperm production, germ cells apoptosis and infertility. Superoxide dismutase (SOD) can eliminate free can prevent oxidative stress and turn back apoptotic process by element free oxygen radical.Because of all these reasons in this study our aim to determine protecting and curative effects of heat stress and SOD which used before stress on testis by using apoptotic and oxidative stress marks.In this study used 18 male Wistar albino, I will be divided into the three groups. Each group contains 6 rats. The first group was identified as a control group of subjects scrotum, temperature 22°C for 20 minutes in the pool were adjusted and testicular tissue were taken 24 hours after the cut. The subjects in the second and third group, in order to create a scrotal hyperthermia, being set in the pool which temperature 42°C for 20 minutes after respectively 30. minutes and 24. hours testicular tissues were obtained per hour. The application of hyperthermia groups enjected with NaCl + SOD + Catalase one hour before application. Tissues which were obtained, compose paraffin blocks which were created by passing methods of light microscopy methods.Testis tissue had been stained caspase 3, 8, 9 and HSP 70 antibody, and we expirimented curative effects of SOD on apoptosis which caused by hypertermia by using immunohistochemical method.After assesment of activator caspase 8, 9, effector 3, we detected that caspase 9 is more markedly. Immunoreactivity increasing specially in spermatogonium, spermatocyte I and Leydig cells by hypertermia exposing. The reason of detecting more caspase 9 positive germ cells, that hypertermia can induce apoptosis by caspase 9 pathway. But caspase 3 did not increase prominently, so we can say that hypertermia did not cause high apoptosis. Consequently we detected that SOD decrease caspase positive germ cells.We detect HSP 70 expression in interstitial location and expression increased with SOD application. This finding indicate of HSP 70?s protecting feature.At the end we can say that hypertermia activate apoptosis specially in spermotogonium, spermatocyte I and Leydig cells through external pathway. Apoptosis decrease testosterone levels in Leydig cells and can be pressed spermatogenesis in seminiferous tubules and can be caused apoptosis through pathway in these cells. But SOD application decrease apoptosis by HSP 70 protective effect.
