Protective effect of purple grape juice on oxidative stress induced by homocysteine

Homocysteine belonging to oxidative stress, is an aminoacid, and due to increase in plasma level, hyperhomocysteinemia takes place. As a result of oxidative stress, reactive oxygen species (ROS) occur and they causes lipid peroxidation and membrane demolitions. There are antioxidant defence system versus ROS harmful effect and antioxidants repairing injuries. Antioxidant-dependent phenolic compounds, such as flavonoids rich enough in purple grape by transmitting a single hydrogen to oxidant make these harmful phenomenas ineffective. This study carried out to determine the effect of grape juice on changes of antioxidant enzyme level in rats for 30 days. 32 Sprague dawley rat were used and study was conducted in four different groups; control, purple grape juice, homocysteine and homocysteine + purple grape juice. Daily normal water and serum physiologic as intra peritonal were given to control group. Daily 10 ml/kg dose grape juice with gavage was given to purple grape juice group. Besides 1 ml/kg serum physiologic was given. 1 mg/kg homocysteine as i.p. and 10 ml/kg grape juice were given to homocysteine + grape juice group. Plasma malondialdehide (MDA), plasma carbonyl, erythrocyte glutation (GSH), plasma sulfhydril (SH), erythrocyte catalaz and erythrocyte superoxide dismutase (SOD) activities were determined. Results revealed that in grape juice group, erythrocyte GSH and erythrocyte CAT levels were found as significantly higher than control group. However, plasma MDA level significantly decreased. In Homocysteine group, plasma MDA and carbonil levels significantly increased and plasma ?SH and erythrocyte SOD levels significantly decreased than control group. In purple grape + homocysteine group, if compered to homocysteine group, plasma ?SH, erythrocyte GSH and erythrocyte CAT levels significantly increased. xv Results as signed that by decreasing MDA level, increasing GSH level and erythrocyte CAT activity grape juice makes decrease oxydative stress induced by homocystein.
