Özet (EN)

Objective: The aim of this study is to ensure that agents that cannot pass through the cell membrane enter the cell with low doses of nanoencapsulation. Material and Methods: For this, hydrogen soy phosphatidylcholine and cholesterol lipids in powder form were purchased as research material. After dissolving these powders in 3: 1 chloroform / methanol, chloroform / methanol was removed under vacuum. Liposome was obtained by homogenizing the toxic chemicals to be encapsulated in the liposome in the obtained lipid layer citrate buffer and passing through the filter. In the last step, this mixture was passed through the column and the liposome was purified from non-encapsulated chemicals and obtained in pure form. Results: The sizes of the resulting liposomes are approximately 200-300 nanometers. The encapsulation efficiency of liposomes loaded with DTPA and TPEN was calculated. The packaging efficiency of DTPA was found to be 43.8%, and the packaging efficiency of TPEN was 1.45%. The IC50 value of DTPA on PC3 cells was 50 µM in 48 hours. The IC50 value of TPEN on PC3 cells was found to be between 4-6 µM in 24 hours. Conclusion: The IC50 value of liposomal DTPA on PC3 cells is evaluated as 50-60 µM in 72 hours. In liposomal TPEN, it was concluded that this value was between 6-8 µM in 72 hours. It was concluded that the toxic effect of free DTPA or TPEN ise more than that of the liposomal formulations.
