INVESTIGATION OF THE PROTECTIVE EFFECT OF HYPERICUM PERFORATUM EXTRACT ON LIPOPOLYSACCARIDE INDUCED CYSTITIS Objective: In our study, we aimed to investigate the protective effect of Hypericum perforatum extract on cystitis induced by lipopolysaccharide. Materials and Methods: In this study, 24 Balb / C strain mice were divided into three equal groups. Intravesical saline was given to the control group transurethrally. Cystitis was induced by giving intravesical LPS to the second and third groups. Before LPS was given to the mice in the third group, daily H. perforatum extract was given by gavage for ten days as a protective. At the end of the experiment, TNF-α, IL-6 levels in serum and iNOS, sFLA2, COX-2, NFκB levels in bladder tissue were measured by ELISA method. One-way analysis of variance (ANOVA) was used for statistical comparisons and Bonferroni was used as a post hoc test. Results: In mice given intravesical LPS, serum IL-6, TNFα levels and inflammatory markers NFκB, COX-2, sFLA2 and iNOS in the bladder tissue were found to be higher than the control group. In the group given H. perforatum extract before LPS, the levels of all these markers were found to be significantly lower. Conclusion: In LPS-induced cystitis, administration of HPE orally before LPS reduced the inflammatory response in the bladder. This study is the first study in the literature investigating the effect of HPE on inflammatory markers in both serum and bladder tissue in LPS-induced cystitis model, and its results may form the basis for studies investigating the place of HPE in the treatment of inflammatory bladder diseases in humans.
