Abstract (EN)

Since ancient times, plants have had a significant place in improving human health. People consider plants safe due to their therapeutic and protective purposes having natural origin and use herbal products for this reason. One of these medically important and traditionally used plants is Hypericum perforatum L. The positive effects of wound healing of this plant, which is widely used to accelerate the healing of burns and wounds among the population, have been proven by studies. In this thesis, soil conditions and genetic variation on essential oil components of H. perforatum L. species belonging to Hypericum genus were investigated. Essential oil components of H. perforatum L. were determined by experimental studies and soil structure was clarified by soil analysis and their genetic characteristics were investigated. Few number of studies on the molecular structure and the comparisons of soil and essential oil components for this plant has led us to do these investigations. Once the H. perforatum L. plant has been collected, it is ensured that the aerial parts are dried in a cool and humid environment. The dried aerial parts was hydrodistilled with a Clevenger device prepared in accordance with European Pharmacopoeia (2010). Volatile oil was obtained after hydrodistillation and its contents were examined by GC/MS. The first stage of analysis at the molecular level is DNA isolation. The DNA of each plant was isolated and prepared for PCR followed by DNA sequencing. Soil color, soil physical analysis, pH, salt in the soil, calcium carbonate, total nitrogen (N), phosphorus, organic matter, iron, copper, zinc and manganese available to plants can be obtained by taking samples of soil 10 cm deep from the place where the plants are collected. Quantity determinations were made. Atomic Absorption Instrument in Eskişehir Soil Research and Ecology Laboratory was used to determine the micro element quantities in the samples.
