Abstract (EN)
Ibuprofen (IBU) is one of the most widely used nonsteroidal anti-inflammatory (NSAI) active substances worldwide. IBU is frequently used in anti inflammatory processes, chronic or acute pain, as well as as an antipyretic in both children and adults. The negative effects of NSAIDs on the duodenum and stomach in the gastrointestinal tract (GIS) have been known for a long time, and studies are still ongoing on this subject. Considering the microbiological and physiological structure of the GIS, there are many difficulties for the dosage form prepared for the oral route to reach the targeted area in the colon without being damaged and to release the active substance. In this thesis, polymeric nanoparticles were prepared by nanoprecipitation method by using Eudragit® S 100 of IBU and polycaprolactone (PCL) polymers with different molecular weights (10000, 45000, 80000) for the colon in order to heal the wounds that may occur in the colon. The in vitro properties of the prepared formulations were examined in detail and the results of the analysis were evaluated. it was concluded that the prepared formulations released the active substance at pH of the colon and the prepared formulations had a wound-healing effect on the cell culture.
