Abstract (EN)
Title: Evaluation of lncRNA GAS5 polymorphism and expression difference in women with idiopathic habitual abortion history Aim: Abortion is defined as the expulsion of all or part of the fetus weighing less than 500 g before the 20th week of pregnancy, and habitual abortion when this event occurs two or more times. Although ~50% of recurrent abortions are idiopathic, causes such as chromosomal anomalies, autoimmune diseases, endocrine diseases, uterine structural anomalies increase the risk of pregnancy loss. In studies, changes in some genetic factors (such as lncRNA GAS5) have been associated with pregnancy loss. In our study, it was planned to investigate the effects of GAS5 gene rs55829688, rs6790, rs2067079 and rs145204276 polymorphisms and GAS5 expression levels on the development of habitual abortion. Method: In our study, DNA and RNA isolation was performed in the samples taken from 75 patients and 75 control groups in order to investigate the relationship between GAS5 gene polymorphisms and expression levels and the risk of pregnancy loss in RPL cases. In the isolated samples, GAS5 rs55829688, rs6790, rs2067079 and rs145204276 polymorphisms were determined by Taq Man SNP and expression level by Real-Time PCR methods. Results: In our study, a statistically significant difference was observed between the groups for the rs55829688 polymorphism (p=0.005), while no difference was found in the genotyping results of the rs6790 and rs2067079 polymorphisms (p=0.241, p=0.086, respectively). No results were obtained from the genotyping study of the rs145204276 polymorphism. In terms of GAS5 expression, the difference between the patient and control groups was significant (p= 0.008). Conclusion: Study data revealed the difference in the level of GAS5 expression between the research and control groups, suggesting that GAS5 may be an RNA associated with pregnancy loss.
