Determination of antibiotic resistance and genetic properties of pathogenic Escherichia coli bacterial strains isolated from urinary tract infection

In collecting 30 urine samples, women (66,66%) were a major source of Uro-Pathogenic E. coli, while men (33,33%) existed. 73,3% of 30 isolates had resistance to two up to thirteenth antibiotics of many classes (even though belonging to the same class). The sensitivity test discloses resistance against trimethoprim+sulphamethoxazole (17) 56,66%, then ciprofloxacin and gentamicin (14) 46,66%. However, (9) 30,00% of ampicillin/sulbactam and cefixime witnessed resistance in 16 antibiotics of different classes. Ertapenem, imipenem, and meropenem all demonstrated little or no resistance, but great sensitivity arose at 93,33; 96,33, and 100%, respectively. Whole genome sequencing (WGS) assembly of five strains incriminated genome length start with 5,24 Gbp for isolate 1; 5,20 Gbp for 2; 5,25 Gbp for 3; 5,25 Gbp for 4 and 5,36 Gbp for 5. For investigating the resistance genes within 5 chosen resistant strains, web-based platform data showed 19 resistance genes belonging to multiple classes: β-Lactamase (blaTEM-1B, blaCTX-M-15, and blaOXA-1) withstand to cephalosporins in assorted generations. sul1, sul2, and dfrA17, dfrA14 donate to trimethoprim+sulphamethoxazole resistance. Aminoglycosides (aph(3")-Ib, aph(3")-Ia, aph(6)-Id, aac(3)IId, aac(3)IIe, aac(6)-Ib-cr, aadA5), tetracyclines (tet(B), tet(A)), chloramphenicol (catA1), fluoroquinolones (qnrS1) and macrolides (mph(A)). Fluoroquinolones overlooked point mutations inwards gyrA, parC, and parE. Virulence factor genes (VFs) were included in this study, isolate 1; 2 and 3 represent (15;13;12) (17,85; 15,47; 14,28%) respectively. Isolate 4 (21) (25%) finally isolate 5 as the highest incidence strain (23) (27,38%). VFs category distributed within our isolates as adhesins 12 genes, iron acquisition system 9, protection and invasion, toxins four, and other miscellaneous proteins 7. Phylogroups were located in four main sorts, isolates one and four located in B1 and B2, respectively, and isolate two, three, and five in the same phylogenetic group D
