Özet (EN)
Today, imaging of biological samples is accepted as a more effective analysis of molecules, cells, and tissues in life sciences. The development of bioimaging technology has enabled researchers to visualize and measure samples in ways that have not previously possible. Quantitative methods in imaging and advances in image analysis reveal the contribution of imaging as a technology to the scientific world. In the thesis study, a bioimaging method has been developed to facilitate the distinction between lung fibroblast cells and lung epithelial carcinoma cells. In the study, the fluorescence iridium complex synthesized by the unique method "AmiNoAcid (monomer) Decorated and Light Underpinning Conjugation Approach" (ANADOLUCA) method has been attached to laponite clay due to its high loading capacity. In addition, immunoglobulin Y (IgY) purified from chicken egg yolk has been used for the antibody orientation. Characterization of the purified IgY has been performed by SDS-PAGE, UV-Vis spectroscopy, FT-IR and CD spectroscopy analysis. The interaction of IgY bound Anti-Transferrin Antibody (ATAb) and transferrin has allowed the complex to enter the cell and imaging on a fluorescence microscope. Characterization studies for Laponite and Iridium (III) (Ir3+) complex have performed by FT-IR, Zeta size analysis, fluorescence spectroscopy, SEM analysis. Finally, the effectiveness of the complex has been demonstrated as a bioimaging agent by imaging with a fluorescence microscope.
