Comparative analysis of cabazitaxel's cellular effects on two- and three-dimensional glioblastoma multiforme (U-87 MG, T98G) cell cultures

There is a common idea about IC50 values of drugs obtained from three dimensional (3D) cell cultures are more in vivo-like than from two dimensional (2D) cell cultures. The objective of this thesis was to investigate and compare the effects of Cabazitaxel (CTX) on cytotoxicity, apoptosis and cell cycle in glioblastoma cell lines [GBM (T98G and U-87MG)] cultured as 2D and 3D. Hanging-drop technique was performed for the formation of 3D (sferoid) cell cultures. Effect of CTX on cell viability was evaluated by using XTT assay and the data obtained from the assay was used for calculating cytotoxicity. The tests which are compatible with Muse Cell Analyzer were used for cell cycle and apoptosis analyzes. IC50 values of CTX were calculated as 1µM (2D T98G, 48 hours), 13.98µM (3D T98G, 72 hours), 9.825µM (2D U-87MG, 72 hours), 92.4µM (3D U-87MG, 96 hours). It was determined that CTX arrested cell cycle at G2/M phase for 2D cultures and G0/G1 phase for 3D cultures. CTX-mediated ealy and late apoptotic cell percentages were determined as 42.75 (2D T98G), 45.56 (3D T98G), 44.36 (2D U-87MG), 27.07 (3D U-87 MG). In this study, it has shown for the first time that cellular effects of CTX differentiate in 2D and 3D GBM cultures. It is suggested in the light of this study's findings that in vitro cytotoxicity studies should perform with not only 2D cell cultures but also with 3D cell cultures for obtaining data in vivo-like.
