Key Words: Ephedrine HCI, Guaifenesin, High Performance Liquid ChromatographyIn this study, the development of a new quantitative determination method with RP-HPLC-DAD (reverse phase-high performance liquid chromatography-Diyod array detector) technique was planned both in pharmaceutical preparations and human plasma for Ephedrine HCI and Guaifenesin active ingredients widely used in cold medicines. For this purpose, simple, rapid, reproducible, sensitive and economical with a DAD (Diyod Array Detector) detector-HPLC (High Performance Liquid Chromatography) method was developed for the quantitative determination as in-vitro in human plasma of a commercial drug.At the same time, analytical method validation process was made after development of analysis both in pharmaceutical preparations and in human plasma without the need for liquid-liquid extraction of this quantitative determination method that could be analyzed simultaneously both active substances. During the validation, all the parameters were determined such as selectivity, specificity, accuracy, recovery, repeatability and the results were evaluated statistically.
