Effect of İL-17, İL-17R, IL-23 and İL-23R gene polymorphisms on the formation of bone destruction at periapical lesions of apical periodontitis patients

Objective: The objective of this clinical study is to investigate the effects of IL-17, IL-17 Receptor (IL-17R), IL-23 and IL-23 Receptor (IL-23R) gene polymorphism on lesion formation in periapical regions of the healthy individuals who have teeth with deep carious lesions and diagnosed with apical periodontitis after the radiographic investigation and examination of the oral cavity. Material and Methods: We included the individuals who diagnosed with deep carious lesions on their teeth after the clinical and radiographic examination between the ages of 18-65 with no systemic illnesses in their medical history. Apical periodontitis patients that have teeth with periapical lesions in their radiographs and gave negative response to thermal and electric pulp tests formed the experimental group; irreversible pulpitis patients that have teeth with healthy periapical tissues on their radiographs and gave positive response to thermal and electric pulp tests formed the control group. Double strand deoxyribonucleic acid (dsDNA) samples which was obtained from the patient's buccal epithelial cells used in polymerase chain reaction (PCR) and also High Resolution Melting analysis (HRM) to determine the genotypes. The ScreenClust HRM (QIAGEN/ Duesseldorf, Germany) software was used to analyze the acquired data to identify the genotypes. Results: There were no statistically significant differences between the apical periodontitis patients and the irreversible pulpitis patients in terms of IL-17A (rs542129933), IL-17RA (rs562543976), IL-23A (rs180825144) ve IL-23R (rs10489630) gene polymorphisms. Conclusion: Within the limitations of this study IL-17 (rs542129933), IL-17R (rs562543976), IL-23 (rs180825144) and IL-23R (rs10489630) single nucleotide polymorphisms (SNP) have no effects on bone resorption in periapical lesions.
