Abstract (EN)
Correlation of immunohistochemically CD144 and Cerb-B2 study results with KRAS, NRAS, BRAF mutations and clinicopathological parameters in advanced stage colorectal carcinoma cases AIM: 50% of deaths in patients with colorectal cancer (CRC) are due to metastases. The main factor in prognosis is TNM stage. However, despite being in the same pathological stage, prognostic differences necessitated understanding the roles of precursor lesions in carcinogenesis. Of these, VE-cadherin is the main molecule in angiogenesis, tumor invasion and metastasis. It is thought to be related to the RAS/RAF and PI3K pathway oncoproteins that regulate epidermal-mesenchymal transition. Revealing this relationship is one of our main goals. MATERIALS AND METHODS: VE-Kaderin expression and Hematoxylin-Eosin sections of 122 metastatic CRC cases diagnosed between 2011-2021 were compared with clinicopathological prognostic parameters. Cases whose blocks could not be reached in the archive and did not contain sufficient tumor tissue were excluded. Cases with large tumor volume, low artifact and low pericolorectal adipose tissue density and at least one of the KRAS, NRAS, BRAF gene mutations were analyzed. Analyzes were made in SPSS program and p <0.05 was considered statistically significant. Log-rank was applied to the cases with Chi-square, Man-whitney-u, Kaplan-Meier methods. RESULTS: Expression of VE-cadherin was observed only in vascular endothelial and tumoral cells in 38 cases. Age, gender, tumor size, type and grade, RAS-RAF positivity, positive KRAS codons, T-N stage, number of lymph node metastases, presence of lymphovascular-perineural invasion did not differ significantly between the groups with and without staining. Between the low and high staining groups, the rate of male patients was found to be significantly higher in the high staining group, and KRAS positivity was found to be lower. CONCLUSION: As in different studies, VE-cadherin was found to play a role in cancer progression. The relationship of VE-cadherin with clinicopathological parameters and KRAS, NRAS, BRAF has not been studied in our country yet, and there are limited studies in the literature. However, further studies are recommended for VE-cadherin-related tumors at different stages. Keys words: HER-2, KRAS, NRAS, vasculogenic mimicry, VE-Kadherin
