Abstract (EN)
In this study, PAL enzyme has been immobilized into gelatin on polyester films. Activity values of free and immobilized PAL enzyme has been found 2,6x10-2 and 6,8x10-3 µmol.min-1, respectively. Specific activity values of free and immobilized PAL enzyme has been found 39,4x10-2 and 13,9x10-2 µmol.min-1 .mg protein-1, respectively. Then immobilized PAL enzyme has been used for analysis of phenylalanine in urine for diagnosis of phenylketonuria. Although immobilized PAL enzyme has been used seventeen times for two months, it has kept its activity. Immobilized PAL enzyme has resulted with less error in high phenylalanine concentration in urine.
