Abstract (EN)
Valeriana officinalis (Valeriana) is an important medicinal plant used as a form of therapy in insomnia, fear, tension and nervousness since ancient times. It is difficult to produce this plant at large quantities due to the small number of seed generation, low germination rate, variations, traditional methods and the cost and time of breeding. Plant tissue culture technique, one of the plant biotechnology techniques, is an alternative way of producing medicinal plants. In our thesis, we aimed to produce V. officinalis plant and its metabolites by plant tissue culture method. The leaves, petioles and cotyledon explants of the germinated plant, in in vitro conditions, were cultured in Murashige and Skoog (MS) nutrient media containing 8 different concentrations and combinations of Benzylaminopurine (BAP) and Naphthalene acetic acid (NAA) plant growth regulators. Metabolites in the roots and calluses were determined by LC-MS method. As a result of the applications, the best root formations in the leaf and leaf extract explants were observed in MS food medium in 10 different concentrations and combinations and best callus formations were observed in 2 mg/L BAP+2 mg/L BAP ve 2 mg/L BAP+1,5 mg/L NAA containing MS culture medium. In addition, no root formation was observed in nutrient media containing 1 mg/L BAP and 2 mg/L BAP.
